Anti-SSA/Ro Antibody Test: Overview

The Anti-SSA/Ro antibody test detects autoantibodies against the SSA (Ro) protein, which plays a role in RNA processing. It is commonly used to diagnose Sjogren’s syndrome, systemic lupus erythematosus (SLE), and neonatal lupus.


Purpose of the Test

Diagnose Sjogren’s syndrome (primary or secondary)
Support the diagnosis of systemic lupus erythematosus (SLE)
Evaluate autoimmune-related dry eyes, dry mouth, or systemic symptoms
Assess pregnancy risk for neonatal lupus & congenital heart block


Interpreting Results

Negative No detectable Anti-SSA/Ro antibodies (does not rule out autoimmune disease)
Low Positive Possible early or mild autoimmune activity, clinical correlation needed
High Positive Strongly associated with Sjogren’s syndrome, SLE, or neonatal lupus

🔹 Anti-SSA/Ro antibodies are found in:

  • 75-90% of Sjogren’s syndrome cases (often with Anti-SSB/La).

  • 30-50% of SLE cases, especially ANA-negative lupus.

  • Neonatal lupus & congenital heart block (CHB) when found in pregnant women.


Clinical Significance

  • Primary Sjogren’s SyndromeHigh Anti-SSA/Ro is a key diagnostic marker.

  • Secondary Sjogren’s Syndrome → Often present alongside rheumatoid arthritis (RA) or lupus (SLE).

  • Lupus (SLE) → Anti-SSA/Ro is linked to photosensitivity, rashes, and ANA-negative lupus.

  • Neonatal Lupus & Congenital Heart Block (CHB) → Pregnant women with Anti-SSA/Ro have an increased risk of fetal heart block (5-10% chance).


Additional Autoimmune Tests

Anti-SSB/La Antibodies → Also common in Sjogren’s and SLE.
ANA (Antinuclear Antibody Test) → Usually positive in autoimmune diseases.
Rheumatoid Factor (RF) → Positive in secondary Sjogren’s syndrome (RA-associated).
Schirmer’s Test & Salivary Gland Biopsy → Confirms Sjogren’s syndrome.
Complement Levels (C3, C4) → Can be low in active lupus.
Fetal Echocardiogram (if pregnant with positive SSA/Ro) → Monitors congenital heart block risk.
